Step-by-Step Sterile Reconstitution Protocol

Sanitize & Prep

Pop off the protective flip-top caps from both the peptide vial and the Bacteriostatic (BAC) water vial. Swab the rubber stoppers thoroughly with a sterile 70% isopropyl alcohol wipe and allow them to air-dry completely (15–30 seconds).

Draw BAC Water

Using a fresh mixing syringe (e.g. 2mL or 3mL with 21G–25G needle), pull an equal volume of room air into the syringe, insert into the BAC water vial, inject the air to balance pressure, and smoothly draw your required water volume (e.g., 2.0 mL).

Slow Wall Injection

Insert the needle through the peptide vial stopper at a 45° angle aimed at the glass wall. Do not squirt water directly onto the lyophilized powder! Allow the vacuum to gently pull the liquid down the inner wall. Control the plunger speed with your thumb.

Swirl & Refrigerate

NEVER SHAKE THE VIAL! Vigorous agitation denatures protein structures. Gently roll the vial between your palms or swirl in slow circular motions until completely dissolved and crystal clear. Store immediately in a refrigerator at 2°C to 8°C (36°F to 46°F).

Understanding Syringe Unit Mathematics

Insulin syringes are calibrated using standard volumetric definitions where 100 Units equals exactly 1.0 milliliter (1 mL):

• 1 Unit = 0.01 mL = 10 µL (microliters)
• 10 Units = 0.10 mL = 100 µL
• 50 Units = 0.50 mL = 500 µL
• 100 Units = 1.00 mL = 1,000 µL

Does a 0.5 mL (50-unit) syringe have different units than a 1.0 mL (100-unit) syringe?
No. 10 units on a 0.5 mL syringe is identical to 10 units on a 1.0 mL syringe (exactly 0.10 mL). The crucial distinction is barrel width: smaller barrels spread tick marks wider apart, providing dramatically higher measurement accuracy and reducing dead space.

Storage Guidelines & BAC Water Lifespan

Peptides exist in two distinct physical states with vastly different degradation profiles:

  • Lyophilized (Dry Freeze-Dried Powder): Stable at room temperature (under 25°C) for several weeks, or in standard freezer (-20°C) for 12 to 36 months protected from light.
  • Reconstituted (Liquid Solution): Must be kept continuously refrigerated at 2°C – 8°C (36°F – 46°F). Do not freeze liquid peptides, as ice crystals can shatter molecular bonds.
  • 0.9% Benzyl Alcohol Window: Bacteriostatic water contains 0.9% benzyl alcohol, which inhibits bacterial proliferation for up to 28 days after first puncture. Solutions older than 30 days should be retired.
